Inhibitory potential of chebulagic acid on digestive enzymes in vitro.

- Abstract
The present study was aimed to investigate the α-amylase, α-glucosidase, and lipase inhibitory effects of chebulagic acid in vitro. Varying concentrations (10, 20, 40, 80, and 100 μg/ml) of chebulagic acid and acarbose were subjected individually to α-amylase and α-glucosidase inhibitory assay, and their absorbance was measured at 540 and 405 nm using a colorimeter. Likewise, the same concentration of chebulagic acid and orlistat was subjected individually to lipase inhibitory assay, and its absorbance was read at 410 nm. Chebulagic acid showed appreciable α-amylase, α-glucosidase, and lipase inhibitory potential in a dose-dependent manner. The maximum percentage of inhibition was observed (68.85 ± 0.28) in α-amylase and (63.35 ± 0.17) in α-glucosidase enzymes at a concentration of 100 μg/ml of chebulagic acid. The standard drug - acarbose - was shown maximum percentage of inhibition in α-amylase (71.46 ± 0.61%) and (69.82 ± 0.66) in α-glucosidase at a concentration of 100 μg/ml. similarly, the highest percentage of inhibition was observed (86.16 ± 0.87) in lipase at a concentration of 100 μg/ml of chebulagic acid. The standard drug orlistat was shown maximum percentage of inhibition (91.61 ± 0.47) at a concentration of 100 μg/ml. The results produced by chebulagic acids were comparable to that of acarbose and orlistat which may be recommended as an oral antidiabetic and antiobese drug.
