Interaction between Phloridzin, Phloretin with DNA Investigated by Electrochemical Probe.

- Abstract
Neutral red (NR) was utilized as a substrate to investigate the interaction of phloridzin and phloretin with herring sperm DNA. A pair of redox peak was obtained in B-R buffer (pH 4.1) with NR and without DNA by cyclic voltammetry. However, we found that the redox current (ip) of NR was decreased markedly by the addition of DNA in solution, thereby causing a reduction of free NR and indicating a binding of neutral red to DNA in solution. Furthermore, we found that addition of phloridzin or phloretin to the reaction mixture of NR and DNA resulted in an increase of redox current (ip), thereby presenting an evidence that NR could be displaced by phloridzin or phloretin which share the same DNA binding site. The competitive binding between drugs and NR was confirmed by absorption spectra. There was a significant difference in DNA binding affinity between phloridzin and phloretin, in which the affinity of phloretin was stronger than that of phloridzin under a same experimental condition.
