Regulatory effects of regulating liver and lung by means of the HPA axis on Thl/Th2 of asthmatic rats with psychological stress.

- Abstract
Objective The changes in serum, lavage fluid and lung tissue were observed in model rats treated with Xiaochuan Ning (Asthma-relieving) Granule to explore the regulatory effects of regulating liver and lung by means of the HPA axis on Thl/Th2 of asthmatic rats with psychological stress. Methods SD rats were randomly divided into the normal group, asthma with psychological stress (APS) group, Pulmicort Respules (PR) group and Xiaochuan Ning Granule (XCNG) group, with 8 rats in each group. The APS group was sensitized with ovalbumin (OVA) and luminum hydroxide adjuvant and stimulated with atomized OVA for 14 days, and restrained simultaneously for 28 consecutive days. The WM group was given atomized Pulmicert Respules 363.11 µG/kg 2 hours before the daily stimulation from the 15 th day of the experiment for 14 consecutive days in addition to the same intervention given to the APS group. The XCNG group was treated with Xiaochuan Ning Granule 2. 18 g/kg by gavage 2 hours before the daily stimulation from the 15th day of the experiment for 14 consecutive days in addition to the same intervention given to the APS group. The normal group was sensitized and stimulated with saline, but was not restrained. Changes in the concentrations of relevant indicators, positive cell expressions and protein expressions in the serum, bronchoalveolar lavage fluid (BALF) and lung tissue were detected with flow cytometry, ELISA, immunohistochemistry and Western blotting to investigate the regulatory effects of regulating liver and lung on the function of hypothalamic pituitary adrenal axis (HPA) and immune inflammation in psychologically stressed asthmatic rats. Results Compared with The APS group, the peripheral blood eosinophil counts were significantly reduced (P <0. 01) and the infiltration of inflammatory cells in lung tissue was significantly reduced of the rats in the XCNG group. The levels of CRH, ACTH and CORT in the serum and BALF of the rats in the XCNG group were significantly lower than those in the APS group (P <0. 01), while the GR positive expressions were significantly higher than that in the APS group (P < 0. 01) though the content was only slightly higher without statistical significance (P >0. 05) . On the other hand, the levels of IL-4 and IL-17 in serum, BALF and bronchial lung tissue were significantly decreased (P <0. 01), and the expression of IFN-γ positive cells in lung tissue was significantly decreased (P < 0. 01) but the content was only slightly decreased without statistical significance (P>0.05). Conclusion Regulating liver and lung may be able to treat asthma with psychological stress by regulating the HPA axis.
