Effects of Acupuncture Combined with Medicine on HSP70 and Local Immunity in Tumor-Bearing BALB/c Mice.

- Abstract
Objective: To study the effects of peritumoral acupuncture combined with cyclophosphamide (CTX) on HSP70 and local immunity in tumor-bearing mice. Methods: The 4T1 breast tumor-bearing mice were randomly divided into a model group, drug group, acupuncture group and group of acupuncture combined with drug. The model group received no intervention; The drug group received intraperitoneal injection with CTX (50 mg/kg) every other day; The acupuncture group received peritumoral acupuncture treatment every day; The group of acupuncture combined with drug received peritumoral acupuncture and intraperitoneal injection with CTX. After 2 weeks of experimental intervention, the inhibition rate was observed. HE staining and IPP6.0 were used to observe the expression of HSP70. Flow cytometry was used to detect the expression of PD-L1 and CD4+CD25+ Treg in tumor tissue. Results: Acupuncture or CTX could inhibit tumor growth and reduce the expression of HSP70. Acupuncture combined with CTX showed synergistic effect on tumor growth and the expression of HSP70. CTX can reduce the expression of CD4+CD25+ Treg cells and increase the positive rate of PD-L1; however, acupuncture or acupuncture combined showed no significant effect. Conclusions: Acupuncture combined with CTX can inhibit the expression of HSP70 in the tumor microenvironment; however, it shows no obvious effect on the expression of PD-L1 and Treg cells.
