- Abstract
Transforming growth factor beta-1 (TGF-β1) plays an important role in regulation of ovarian functions. In the present study, it was investigated whether low dose capsaicin (CAP) administration in rat ovaries was effected on TGF-β1 expression during folliculogenesis and atresia. Thirty 21-day-old female Sprague-Dawley rats were divided randomly into three groups. The first group received no treatment (control A group), the second group received (0.5 mg/kg/day) CAP dissolved in the vehicle (experimental group), and the third group was treated with the (0.1 mg/kg/day) vehicle only (control B group). After 15 days treatment period, animals were euthanized by ether inhalation and ovarian tissues were sampled and processed routinely. TGF-β1 was demonstrated by labeled streptavidin-biotin technique. Oocytes, granulosa cells of all follicles (primordial, primer, secondary, preantral and antral follicles), theca cells and interstitial cells were stained positively in experimental group. In control group, the staining was generally in similar fashion but there was no staining in granulosa cells of preantral and antral follicles. TGF-β1 staining was more intense in experimental group than both control groups. There was no TGF- β1 staining in experimental group and control groups' atretic follicles. As a result, we demonstrated that binding of CAP to its receptor in ovarian cells resulted with increased TGF-β1expression during the folliculogenesis.
