- Abstract
A high-performance thin-layer chromatographic (HPTLC) method has been proposed for the analysis of quercetin in Ocimum sanctum by recording peak areas at 365 nm. The correlation between the concentration and area was found to be linear within the range of 100 to 600 ng/spot. The method was validated for precision (interday and intraday), repeatability, and accuracy. Mean recovery for quercetin was 98.88-100.58%. The method was applied for the quantitation of quercetin in Tulsi and found to be 0.74 mg/mL of extract. The proposed HPTLC method was found to be precise, specific, sensitive, and accurate and can be used for routine analysis of Tulsi. The total phenolic and flavonoid content of Ocimum sanctum is 82.02 ± 8.17 mg GAE/g and 74.6±5.1 mg/g respectively.
