Enrichment of Gut Ecosystem by Daily Supplementation of Selective Probiotic Strains and Probiotic Complex in Dysbiosis Condition of Autism.

- Abstract
Context: Probiotics are functional foods involved in gut health support and establishment of Gut-Brain-Axis, adequate supplementation of probiotics needed in autism. Background: Gut microbial ecosystem is very essential for the functioning of gut and establishment of proper functioning between gut and brain via gut-brain-axis. This article aims to evaluate the effect of selected probiotic strains supplementation and probiotic complex on the microbiota colonization in the gut and recognize its implication in autism. Dysbiosis is important comorbid condition of autism. Daily administration of probiotics can impact on microbiota composition and also improves intestinal performance, which is useful for reversing of behavioral abnormalities in autism. Method: On embryonic day 12, valproic acid was administered at a dosage 400 mg/kg, i.p. the born rats exhibited autistic features, treatment duration for this study is postnatal day (PND 8-50) with the Selective probiotic strains and probiotic complex with a dosage Not Less Than (NLT) 1 Billion (CFU) Colony Forming Units CFU/ml, p. o. Results: Selective probiotic strains and Probiotic complex treatment significantly ameliorated in behaviors (p<0.001) Negative geotaxis (15.60±0.67 vs 1.80±0.20), T-maze (0.714±0.184 vs 4.00±0.00), Morris water maze (111.8±7.81 vs 12.66±1.43), Social interaction (6.83±0.74 vs 23.33±0.66), Repetitive/Stereotypic like behavior (7.40±0.67 vs 13.80±0.86), increased biochemical were significantly attenuated IL-6 (47.58±1.07 vs 27.45±0.57), TNF-a (134.61±1.85 vs 100.1±1.56), BDNF (59.74±1.07 vs 45.57±1.06), 5-HT (26.84±1.31 vs 8.39±1.74), AchE (6.40±0.36 vs 1.64± 0.12) along with restoration of areas of cerebellum Molecular layer and Granular layer become normal. Conclusion: Daily Probiotics supplementation to the dysbiotic autistic rats can enriches the microbial diversity and colonization of gut with useful microbiota, that can alter the conditions of Dysbiosis and improves the functioning capability of Gut-Brain-Axis. In our study with the Probiotics treatment it is understand that supplementation of Probiotics and its complex is needed in reversing of Dysbiosis, also alters the behavioral, biochemical and histological abnormalities in autism.
