- Abstract
A liquid chromatography tandem mass spectrometry (LC-MS/MS) method to determine etoricoxib in human plasma is needed for bioequivalence and pharmacokinetic studies of etoricoxib preparations. The present study describes a simple, rapid, reproducible, and reliable LC-MS/MS method to determine etoricoxib concentrations in human plasma. After one-step liquid-liquid extraction (LLE) using methyl tert-butyl ether (MTBE), etoricoxib and etoricoxib D3 (internal standard, IS) were eluted on a Thermo Hypurity, C18 column with an binary mobile phase, consisting of 2mM ammonium acetate buffer: methanol (20:80, v/v) at a flow rate of 1.2mL/min. The achieved lower limit of quantitation (LLOQ) was 19.670 ng/mL and the standard calibration curve for etoricoxib was linear (correlation coefficients were >0.98) over the studied concentration range (19.670-6553.297 ng/mL). The chromatographic run time for each plasma sample was 3.50 min. The developed method was successfully applied to a bioequivalence study of etoricoxib in healthy male volunteers.
