- Abstract
Objective To explore the mechanism of LncRNA TUG1 in hypertrophic scar. Methods From December 2019 to December 2022,Department of Burn and Plastic Surgery, General Hospital of Northern Theater Command, the expression of TUG1 in hypertrophic scar tissue and normal skin tissue was detected by gene biochip technique and verified by Real-time PCR. Fibroblasts (FBs) extracted from HS were divided into two groups and transfected into si-NC and si-TUG1 respectively. The expression of TUG1 was detected by Real-time PCR. Western blot assay was used to detect the expression level of MMP2 in FBs after downregulation of TUG1. MTT assay was used to detect the cell viability at 0, 12, 24, 36, 48 h after transfection, and Transwell assay was used to detect the migration ability of FBS in the two groups. Results The expression content of TUG1 in HS was (1.2600±0.1231), which was significantly higher than that in adjacent normal tissues (0.7052±0.1035) (t=3.447, P<0.05). After down-regulating the expression of TUG1 in HFs (t=20.48, P<0.05), the results of the MTT assay showed that the proliferation rate of FBs in the down-regulated TUG1 group was lower than that in the control group, and the difference was statistically significant (t=3.637, P<0.05). The results of Transwell showed that the ability of FBs migration in the down-regulated TUG1 group was significantly lower than that in the control group (t=12.98, P<0.05). The results of the Western blot showed that after downregulating TUG1, the expression level of MMP2 in FBs decreased. Conclusion The expression of TUG1 is up-regulated in HS and promotes the biological process of FBs by promoting the proliferation and migration of HS.
