- Abstract
pH imbalance has been implicated in various diseases such as cancer and neurodegeneration. However, the underlying mechanisms remain unknown due to a lack of proper tools for pH manipulation in subcellular locales, specific cell types, and tissues. In our recent study, we have introduced Chemogenetic Operation of iNTRacellular prOton Levels (pH-Control) approach, enabling pH manipulation with high spatiotemporal resolution. We have fully characterized pHControl in vitro and in cellulo. Combining pH-Control with genetically encoded pH biosensors (SypHer3s) in HEK293 cells we have shown that pH-Control is capable of changing pH in subcellular locales such as mitochondria, nucleus, and cytoplasm. Besides, we have shown that the activity of pH-Control in dorsal root ganglion (DRG) neurons decreases the membrane potential. Overall, we are convinced that pH-Control has the potential to open new lines of investigation on the role of pH in health and disease.
